Effects of Chiljehyangbuhwan on growth of human uterine cell lines, ovulation and on the early embryonic development

Abstract
Purpose
These experiments were carried out to evaluate of Chiljehyangbuhwan as a female myoma or cancer treatment medicine on in vitro growth of three different uterine cell lines and ovulation rate and in vitro preimplantation embryonic development following Chiljehyangbuhwan administration in the mouse.
Method
The experimental groups are Rhizoma cyperi (Sample ¥°) and Chiljehyangbuhwan (Sample ¥±) group. In first experiment, In vitro growth rate and viability of three different uterus derived cell lines, human uterine normal cell line, human uterine myomal cell line and human uterine adenocarcinormal cell line, culturing in media containing Rhizoma cyperi and Chiljehyangbuhwan for 3 days were compared. In second experiment, total ovulated oocytes from superovulated mice and in vitro embryonic developmental rate from zygote to hatched blastocyst stage were investigate in Rhizoma cyperi or Chiljehyangbuhwan administrated mice.
Results
1. In vitro growth rate was significantly lower in uterine myomal cell line and uterine adenocarcinormal cell line than uterine normal cell line and viability was also decreased in uterine myomal cell line and human adenocarcinormal cell line culture in media containing Rhizoma cyperi and Chiljehyangbuhwan.
2. Minimal concentration showing growth inhibitory action was observed 0.1% of uterine myomal cell line and 2.5% of uterine adenocarcinormal cell line in Rhizoma cyperi, and 1% of uterine myomal cell line and 5% of uterine adenocarcinormal cell line in Chiliehyangbuhwan, respectively.
3. The viability of cultured uterine myomal cell line and uterine adenocarcinormal cell line was showed dose-dependent manner. Also, viability of uterine myomal cell line was lower than that of uterine adenocarcinormal cell line, and viability of cell lines cultured in Rhizoma cyperi was lower than that of Chiljehyangbuhwan
4. Ovulation rate in the mouse administrated Chiljehyangbuhwan by 4 days was significantly decreased than those of control group and Rhizoma cyperi group, respectively.
5. Early embryonic development rate was significantly decreased in both treatment groups, Rhizoma cyperi and Chiljehyangbuhwan groups, than that of control group.
